Abstract: Poly(ethylene glycol) (PEG) lipids are used in lipid nanoparticles (LNPs), i.e. for Covid-19 vaccination. A concern of PEG is the formation of anti-PEG antibodies (APAs) that can cause loss of drug-functionality and side effects. A new class of lipids containing randomized PEG (rPEG) was established. A set of rPEG copolymers was synthesized and conjugated to lipid anchors. Interaction of rPEG lipids with APAs was quantified by enzyme-linked immunosorbent assay (ELISA). rPEG lipids were formulated to LNPs and tested in vitro. Target molecular weight and Dispersity (D95%) of rPEG lipids confirmed successful conjugation. ELISA confirmed significantly reduced interaction with APAs. rPEG LNPs had defined and controllable diameters (100-140 nm) and were found fully functional in vitro. The new class of rPEG lipids is applicable to industrial manufacturing processes and a candidate to fuel next generation mRNA therapeutics.
